Verve Therapeutics - $46,996,500 </h3>On June 21, chief scientific officer Andrew Bellinger bought 5,000 shares, totaling $95,000. On the same day, CEO Sekar Kathiresan bought 2,500 shares amounting to $47,500 and vice president of finance Margaret Beaudoin bought 1,000 at a value of $19,000.As for 10% owners, Casdin Partners Master Fund bought 1.1 million shares to total $20.9 million, while Logos Global Management and Rock Springs Capital each bought 370,000 shares, amounting to around $7 million apiece.Novo Holdings bought 50,000 to total $6.7 million, while and Cormorant Private Healthcare Fund bought 275,000 to amount to $5.2 million. <ul class="summary-list"><li>Ark Invest's flagship ETF is sitting on massive losses, with individual holdings down as much as 93%.</li><li>The fund has significantly underperformed the broader stock market, and only one of its top 10 holdings is in the green.</li></ul>It's been <a href="https://markets.businessinsider.com/news/funds/cathie-wood-ark-invest-wealth-destroyed-tech-stocks-morningstar-arkk-2024-2">a tough stretch for Cathie Wood and her suite of ARK Invest ETFs.</a>Ark Invest's <a href="https://markets.businessinsider.com/etfs/ark-innovation-etf-us00214q1040?utm_medium=ingest&amp;utm_source=markets&amp;utm_medium=ingest&amp;utm_source=markets">flagship innovation ETF,</a> which trades under the ticker symbol "ARKK," <a href="https://markets.businessinsider.com/news/funds/cathie-wood-ark-invest-wealth-destroyed-tech-stocks-morningstar-arkk-2024-2">has seen massive losses since it peaked in February 2021.</a>The ARKK ETF is down 72% since its February 19, 2021 peak. Over the same time period, the Nasdaq 100, its primary benchmark, has soared 39% and it sitting at record highs. <ul class="summary-list"><li>A 13-year-old girl was the world's first patient to get a cell therapy called base editing in May.</li><li>The experimental treatment has put her leukemia in remission for six months and counting.</li></ul>A British teenager's leukemia went into remission after she received an experimental cancer therapy that used a new gene-editing technology called base editing.Alyssa, 13, was the first-ever patient to receive a base-edited cell therapy after enrolling in a clinical trial in May. A month after the infusion and a subsequent bone-marrow transplant, her cancer, called T-cell acute lymphoblastic leukemia, went into remission, and the Leicester resident remains in remission six months later, the Great Ormond Street Hospital for Children <a href="https://www.gosh.nhs.uk/news/gosh-patient-receives-world-first-treatment-for-her-incurable-t-cell-leukaemia/">said on December 11</a>. The dramatic success is an early sign of the potential of base editing, a new technology discovered <a href="https://www.businessinsider.com/how-base-editing-could-change-future-gene-editing-and-crispr-2022-2">less than a decade ago in a Harvard University laboratory</a>. Base editing allows scientists to make ultraprecise changes to single letters of DNA in cells. The goal is to make genetic tweaks that could treat or cure diseases.Beyond leukemia, biotech companies are aiming to use base-editing therapies to <a href="https://www.globenewswire.com/news-release/2022/11/14/2554716/0/en/Beam-Therapeutics-Enrolls-First-Patient-in-BEACON-Clinical-Trial-of-BEAM-101-Base-Editing-Therapy-Candidate-for-the-Treatment-of-Sickle-Cell-Disease.html">treat sickle-cell disease</a> and <a href="https://ir.vervetx.com/news-releases/news-release-details/verve-therapeutics-doses-first-human-investigational-vivo-base">some types of cardiovascular disease</a>.<div type="image" title="Alyssa, a teenager in Leicester, England, received a base-edited cell therapy to treat her leukemia." height="1333" width="1000" msid="96351688" hostid="261"></div>In the case of Alyssa, her treatment used base editing to genetically edit T-cells, a type of immune cell, that came from a healthy donor. These cells were edited to allow them to destroy cancerous cells while not attacking her own immune system, and then infused into Alyssa."It's our most sophisticated cell engineering so far and paves the way for other new treatments and ultimately better futures for sick children," Dr. Waseem Qasim, a cell- and gene-therapy professor and consultant immunologist at Great Ormond Street Hospital, <a href="https://www.gosh.nhs.uk/news/gosh-patient-receives-world-first-treatment-for-her-incurable-t-cell-leukaemia/">said in a statement</a>. <h2>Base-editing is an ultraprecise version of CRISPR gene editing</h2>Base editing was discovered in the lab of the Harvard genomics researcher David Liu.In November 2013, a 26-year-old chemist named Alexis Komor exchanged emails with Liu to figure out a research project, <a href="https://www.businessinsider.com/how-base-editing-could-change-future-gene-editing-and-crispr-2022-2">as Insider has reported</a>. Komor and Liu sketched out the base-editing idea in just a few days, which sparked years of research that culminated in their results being published in April 2016 in <a href="https://affiliate.insider.com/?postID=6214fdaae1ca0a2c043668ea&amp;site=bi&amp;u=https%3A%2F%2Fwww.nature.com%2Farticles%2Fnature17946&amp;amazonTrackingID=null&amp;vikingID=6214fdaae1ca0a2c043668ea&amp;platform=browser&amp;sc=false&amp;disabled=false&amp;sessionID=1671049791095iznteh4h" rel="nofollow sponsored noindex"> Nature</a>, a top scientific journal.<div type="image" title="A headshot of Alexis Komor, a developer of base editing and genomics researcher at University of California, San Diego" height="1985" width="2647" msid="96351689" hostid="261"></div>That paper has now been cited over 3,400 times and has helped <a href="https://www.businessinsider.com/list-top-crispr-and-gene-editing-biotechs-to-know-in-2022-2022-1">launch multiple biotech companies,</a> including Beam Therapeutics, cofounded by Liu, and Verve Therapeutics."It's really crazy that this Frankenstein genome-editing tool put together from all these different parts works so well," Komor <a href="https://www.businessinsider.com/how-base-editing-could-change-future-gene-editing-and-crispr-2022-2">previously told Insider.</a>The technology addresses some of the <a href="https://www.businessinsider.com/crispr-outlook-for-2022-top-gene-editing-experts-2021-11">limitations of CRISPR-Cas9</a>, the Nobel Prize-winning gene-editing technology discovered in 2012. CRISPR-Cas9 works by cutting a disease-causing gene out of the DNA double helix before stitching it back up. Instead of making a full cut, base editors nick a single strand of DNA, while simultaneously changing a single letter of DNA on the other strand. The cell then repairs that nick and uses the just-edited DNA as its template.The technology gives researchers an unprecedented level of specificity, being able to make changes to the base elements of genetic code, or the nucleotides nicknamed A, C, G, and T. Researchers often compare base editing to using a pencil and eraser, while the traditional CRISPR system is more like using scissors and glue.Despite these advantages, base editing also has its limits. Base editors can only change a single letter of genetic code, and researchers haven't yet figured out how to make all the changes. As of now, base editors can make only <a href="https://affiliate.insider.com?postID=639a2dd771518a1580564597&amp;site=bi&amp;u=https%3A%2F%2Fwww.nature.com%2Farticles%2Fd41586-022-01117-z">certain changes to nucleotides</a>, limiting its potential. For instance, base editing can change an A to a G but not an A to a T. Newer technology, <a href="https://affiliate.insider.com/?postID=6214fdaae1ca0a2c043668ea&amp;site=bi&amp;u=https%3A%2F%2Fwww.nature.com%2Farticles%2Fs41434-021-00263-9&amp;amazonTrackingID=null&amp;vikingID=6214fdaae1ca0a2c043668ea&amp;platform=browser&amp;sc=false&amp;disabled=false&amp;sessionID=1671049709382rxjb32m5" rel="nofollow sponsored noindex">like prime editing</a>, can write and edit longer stretches of letters.<h2>Big Pharma sees 'tremendous potential' in base editing</h2>The potential of base editing doesn't stop with Alyssa, as researchers are starting to test base-editing therapies in a range of conditions. Earlier this month, the FDA <a href="https://www.globenewswire.com/news-release/2022/12/02/2566549/0/en/Beam-Therapeutics-Announces-FDA-Has-Lifted-the-Clinical-Hold-on-the-Investigational-New-Drug-Application-for-BEAM-201.html">cleared a different T-cell therapy that is similar to Alyssa's treatment</a> to start initial human testing."Seeing more patients like Alyssa experience remission from this treatment would mean so much both to the researchers who developed base editing, and to the communities of patients who might benefit," Liu told Insider in an email.Big Pharma has also shown interest, with Pfizer <a href="https://www.businessinsider.com/pfizer-enters-gene-editing-pact-with-beam-therapeutics-2022-1">agreeing in January 2022 to pay $300 million</a> up front to collaborate with Beam on several base-editing therapies. Mikael Dolsten, Pfizer's chief scientific officer, told Insider at the time he anticipates base editing having an impact for large patient populations over time — and not just for rare diseases."This is a curative technology and can really transform genetic medicine for the future," Dolsten said. "It has tremendous potential."

<a href="https://businessinsider.com/100-people-transforming-business-2021-11" target="_blank" rel="noopener"></a><div type="image" title="David Liu" height="2354" width="3139" msid="87853929" hostid="261"></div><ul class="summary-list"><li>Scientist and <keyword id="11908912" type="UNKNOWN" weightage="60" keywordseo="biotech" source="Orion">biotech</keyword> cofounder <keyword id="12586351" type="UNKNOWN" weightage="60" keywordseo="david-liu" source="Orion">David Liu</keyword> is one of the pioneers of drugs that can edit DNA.</li><li>His lab is developing therapy that could prevent Alzheimer's by installing a protective gene.</li></ul>David Liu <a href="https://www.businessinsider.com/100-people-transforming-business-2021?person=david-liu-healthcare">is best known</a> as one of the pioneers of the experimental science of editing people's genes.Liu's laboratory at the Broad Institute of MIT and Harvard was the birthplace of two new types of gene-editing called base and prime editing, that swap out segments of the DNA strand without cutting into it. He and other experts estimate could be more precise and have less unintended effects than other forms of gene-editing like CRISPR. What's less known is that for the last five years, his laboratory has been quietly testing a method of potentially protecting people against one of the <a href="https://www.who.int/news-room/fact-sheets/detail/dementia">most devastating health conditions worldwide</a>: <keyword id="11948157" type="UNKNOWN" weightage="60" keywordseo="alzheimers-disease" source="Orion">Alzheimer's disease</keyword>. One of the lab's first tests of this science is to see if it can be used to install a gene called APOE2 that is believed to significantly reduce the risk of a person getting Alzheimer's. The work is still in the very early stages, Liu told Insider. His team has <a href="https://www.nature.com/articles/nature17946">published</a> <a href="https://www.nature.com/articles/nature17946">multiple papers</a> indicating that they can make these changes in cell samples and is currently running additional experiments on mice. The concept of preventing Alzheimer's is gaining traction, with I-Mab Biopharma launching an early-stage trial earlier this week of a <a href="https://www.bostonglobe.com/2021/11/16/business/brigham-womens-hospital-is-testing-nasal-vaccine-alzheimers/">nasal vaccine for the disease</a>. Biotechs <a href="https://www.biospace.com/article/is-a-vaccine-for-alzheimer-s-on-the-horizon-/">Nuravax</a> and <a href="https://medcitynews.com/2021/11/with-claims-of-potential-alzheimers-edge-vaxxinity-squeaks-out-88m-ipo/">Vaxxinity</a> are also working on Alzheimer's vaccines. The benefits of gene-editing is that it could be a one-and-done drug. But it's still highly experimental, and is only being tested in humans with rare diseases right now. There's still many questions to be answered before tweaking the DNA of the 33 million people worldwide with Alzheimer's disease, let alone giving a new type of medication to people who are otherwise healthy but at risk of Alzheimer's disease in the future. Liu's Alzheimer's work is not only a significant test of new science — it could also help determine if and how gene-editing goes mainstream. <h2>Liu's work focuses on a gene that makes people up to 99.6% less likely to get Alzheimer's</h2>No one knows exactly what causes Alzheimer's disease. At this point, there are a <a href="https://www.businessinsider.com/alzheimers-disease-scientists-new-approaches-treat-2021-3">myriad of theories</a> because there's not many biological characteristics that are found in every person with the disease. Most drug companies have focused on a protein called amyloid beta that builds up in the brain of some Alzheimer's patients, but not all patients have that buildup. There is a genetic mutation that has strong links to the disease: the APOE4 gene. But that, too, isn't definitive — just because you have the mutation doesn't mean you will get Alzheimer's disease.However, there may be another way to stop the disease.Research has shown that people with a similar gene, APOE2, are <a href="https://www.nature.com/articles/s41467-019-14279-8">between 66% and 99.6% less likely</a> to develop Alzheimer's disease. Liu and his research team zeroed in on this gene as they were <a href="https://www.genengnews.com/insights/all-about-that-base-editing/">developing the first base editing technology</a> between 2013 and 2015. It was a somewhat arbitrary idea initially, Liu said, but checked all of the boxes: There was strong science linking APOE2 to Alzheimer's disease and promising signs that base editing could tweak that gene. The idea is to develop a treatment that would flip the baseline letters of the DNA strand — G, A, T and C — to turn APOE4 or another type of gene known as APOE3 into APOE2.The lab is still running tests to answer key questions like how and where in the body to deliver the treatment, according to Liu.<h2>Experts still wonder if gene therapies are too risky for the general population</h2>Gladstone Institutes Associate Investigator Yadong Huang, who has studied APOE4 since 1985, thinks that Liu's idea of using gene-editing is the right strategy. It's clear that APOE2 protects against developing Alzheimer's disease, and Huang likes that the Liu lab's approach doesn't involve cutting the DNA strand, as scientists theorized that cutting the strand could cause problems. It does raise many questions, though. Who should get it: everyone, or only people with the high-risk APOE4 gene, asked neurologist-turned-venture-capitalist Doug Cole. And when would they need to get the <keyword id="12586350" type="UNKNOWN" weightage="60" keywordseo="gene-therapy" source="Orion">gene therapy</keyword>? Liu and his research team haven't answered those questions yet. But Liu pointed to another startup he co-founded, Beam Therapeutics, which is <a href="https://www.vervetx.com/press-releases/verve-therapeutics-presents-new-data-in-non-human-primates-validating-gene-editing-as-a-treatment-approach-for-coronary-heart-disease-at-the-isscr-2020-virtual-annual-meeting/">working with Verve Therapeutics</a> on a gene therapy that could <a href="https://www.businessinsider.com/verve-ceo-interview-gene-editing-for-heart-disease-new-data-2021-1">potentially cure heart disease</a>. They're starting with patients who have a high risk of heart disease due to a genetic mutation, and stand to benefit the most from treatment."All of these therapeutics boil down to how to best balance potential benefit to the patient, to the potential risk to the patient," Liu said. "We have a whole plethora of next-generation CRISPR and non-CRISPR gene-editing technologies that offer new capabilities and could potentially be much more broadly applicable."
