New Delhi, Sep 25 (IANS) The Post Graduate Institute of Medical Education and Research (PGIMER), Chandigarh, has administered the first dose of Oxford Covid-19 vaccine -- Covishield -- to three candidates on Friday, informed PGI Chandigarh Director Prof Jagat Ram. The clinical trials of Oxford vaccine has started at PGI Chandigarh after the institute administered first dosage of the vaccine to three volunteers on Friday, Prof Jagat Ram, director, PGIMER told IANS. Ram said that two females and a male were given dosage of 0.5 ml. "Two females aged 57 and 26 years and a 33-year-old male were administered the vaccine candidate at the institute," he specified. The PGIMER is one of the seventeen sites where phase II trials of the 'Covishield' vaccine developed by Oxford University and being mass-produced by Pune based Serum Institute of India (SII) is being conducted. A source privy to the development confirmed that the candidates will be monitored for 28 days after the 0.5 ml dose of vaccine and then the second dose will be given followed by a monitoring of 28 days. After that a blood sample will be taken and they will be observed for a span of six months. Besides, the candidates have been asked to note any development of symptoms like fever in their personal diary. 18 candidates have been found eligible till now to undergo the trial. The institute plans to take 100 candidates into the phase II study while it has made commitment for 253 candidates to Indian Council of Medical Research (ICMR). The criteria fulfilling the eligibility prerequisites for the study included the minimum age of 18, no underlying medical condition and no prior Covid-19 infection. Besides, family members of the candidates are also required to be Covid negative. The trials started after the institute received safety approval pending from the Data Safety and Monitoring Board (DSMB) for the first 100 candidates selected to undergo the trials last week. The trails have been delayed by more than two weeks. The institute had earlier claimed to start the trials by the first week of September. However, it was initially delayed due to pending safety approvals of the candidates from DSMB. Later, the trials were suspended pan-India by Central Drugs Standard Control Organisation (CDSCO) after COVID-19 vaccine trials were put on hold in the United Kingdom (UK) as one of the participants there reported a severe adverse reaction following administration of a booster dose. PGI Chandigarh starts Oxford Covid vaccine trial

London, Dec 9 (IANS) In a significant development, the first full results from interim analysis has confirmed that the Oxford Covid-19 vaccine has an acceptable safety profile and is efficacious against symptomatic the Coronavirus caused disease, with only three out of 23,745 participants experiencing serious adverse events. Interim results of the Oxford Covid-19 vaccine trials found that the vaccine protects against symptomatic disease in 70 per cent of cases -- with vaccine efficacy of 62 per cent for those given two full doses, and of 90 per cent in those given a half then a full dose. The results are the first full peer-reviewed efficacy results to be published for a COVID-19 vaccine, and are published in The Lancet. The results were based on a pre-specified pooled analysis of phase 3 trials in UK and Brazil (11,636 people), alongside safety data from a total of 23,745 participants in 4 trials in the UK, Brazil and South Africa. All participants have recovered or are recovering, and remain in the trial. "The results presented in this report provide the key findings from our first interim analysis," said study author and doctor Merryn Voysey from University of Oxford. "In future analyses, with more data included as it becomes available, we will investigate differences in key subgroups such as older adults, various ethnicities, doses, timing of booster vaccines, and we will determine which immune responses equate to protection from infection or disease," the author said. The Oxford Covid-19 vaccine uses a chimpanzee adenovirus viral vector that cannot cause disease in humans and expresses the SARS-CoV-2 spike protein. This means the vaccine delivers the spike protein genetic code into vaccinated people's cells, which then produce the protein, and teaching the immune system to recognise and attack the virus. Past trial results have found that the vaccine induces antibody and T cell immune responses, and is safe in adults aged 18 years and over, including older adults, the study noted. "Our findings indicate that our vaccine's efficacy exceeds the thresholds set by health authorities and may have a potential public health impact," said study lead author Professor Andrew Pollard. Cases of severe disease and hospitalisation were monitored for in all 23,745 participants. From 21 days after the first dose there were 10 cases hospitalised for Covid-19, all in the control arm, and two were classified as severe, including one death. "We have shown for the first time that an adenoviral vectored vaccine - a type of vaccine technology which has been in use since 2009 - is efficacious and could contribute to disease control in the Covid-19 pandemic," said Professor Sarah Gilbert from University of Oxford. The authors noted that they are not yet able to assess duration of protection, as the first trials were initiated in April 2020 and all disease episodes have accrued within six months of the first dose being administered. He emphasized that the WHO's Global Advisory Committee on Vaccine Safety has been reviewing the available data, is in close contact with the European Medicines Agency (EMA) and will meet on Tuesday. AstraZeneca vaccine is still on WHO's radar

New Delhi, Sep 6 (IANS) The clinical trials for the Covid-19 vaccine candidate developed by the University of Oxford, which was expected to start at The Post Graduate Institute of Medical Education and Research (PGIMER) in Chandigarh, have been delayed by at least a week. The trials that were expected to commence in the first week of September have been delayed due to the safety approval pending from the Data Safety and Monitoring Board (DSMB) for the first 100 candidates selected to undergo the trials, the institute told IANS. In result, the further recruitment of the candidates for the trials has been put on hold. "For the time being, the further recruitment of the candidates for the trials for the Oxford vaccine is on hold because we are waiting for the approval for safety of the first 100 participants recruited till now from the Data Safety and Monitoring Board. In this light, we would be able to update you regarding any development on this front by next weekend only," said Prof. Dr. Madhu Gupta, the principal investigator of the vaccine trial scheduled at the PGIMER. The premier institute said it received 400 volunteers for the trials. Of them, 253 would be administered the first dose of the undertrial vaccine. Gupta had earlier informed that 16-member staff, including herself, would supervise the clinical trials. The PGIMER is among the 17 sites selected for the human clinical trials of the adenovirus-based Covid-19 vaccine candidate developed jointly by AstraZeneca and the University of Oxford. India. Oxford Covid vaccine trials delayed over safety approvals

As the country awaits the UK's approval to the AstraZeneca-Oxford vaccine before giving nod for Emergency Use Authorisation to its Indian counterpart developed by Pune-based <keyword id="95879" type="ORGANIZATION" weightage="20" keywordseo="Serum-Institute-of-India">Serum Institute of India</keyword> (SII), a top European drug official has claimed that the vaccine is not even ready for a conditional approval at the moment. Noel Wathion, number two at the European Medicines Agency, told Belgian newspaper Het Nieuwsblad on Tuesday that the approval for AstraZeneca-Oxford vaccine is still "unthinkable" and Europe is not going to approve the vaccine with "winning formula" for the time being. "They haven't even submitted an application to us yet," Wathion was quoted as saying in the report. The UK aims to start rolling out the British AstraZeneca/Oxford vaccine from January 4. According to Wathion, "The data we have at the moment is not even enough to give the AstraZeneca vaccine a conditional authorisation". "We need additional data about the quality of the vaccine. And after that, the company has to formally apply," Wathion added. Pune-based Serum Institute of India on Monday expressed hope at the possibility of its vaccine called Covishield receiving the regulatory approval in "few days", while emphasising that the company has a massive stockpile of 40-50 million doses. "We have 40-50 million doses of Covishield stockpiled. Once we get regulatory approvals in a few days, it will be down to the government to decide how much they can take and how fast. We will be producing around 300 million doses by July 2021," Serum Institute CEO Adar Poonawalla said during a press conference. India currently has eight Covid-19 vaccine candidates, including three indigenous vaccines, under different stages of clinical trials which could be ready for authorisation in near future. Serum Institute-Oxford's Covishield, Bharat Biotech's Covaxin and Pfizer vaccine are in the fray for emergency use authorisation. The central government plans to vaccinate nearly 30 crore people in the first phase of drive. It will be offered to one crore healthcare workers, along with 2 crore frontline and essential workers and 27 crore elderly, mostly above the age of 50 years with comorbidities. Oxford-AstraZeneca COVID-19 vaccine approval is 'unthinkable', claims European watchdog

Serum Institute of India to soon apply for local trials of AstraZeneca-Oxford Covid-19 vaccine

Once we get regulatory approvals in a few days, it will be down to the government to decide how much they can take and how fast. We will be producing around 300 million doses by July 2021," said Serum Institute CEO Adar Poonawalla during a press conference on Monday. However, officials at the European Medicines Agency (<a href="https://www.businessinsider.in/science/health/news/oxford-astrazeneca-covid-19-vaccine-approval-is-unthinkable-claims-european-watchdog/articleshow/80022921.cms">EMA</a>) have warned that giving the vaccine approval at this stage is ‘unthinkable’. "The data we have at the moment is not even enough to give the AstraZeneca vaccine a conditional authorisation," they told Belgian newspaper het Nieuwsblad. "We need additional data about the quality of the vaccine. And after that, the company has to formally apply.” As of now, AstraZeneca recommends two doses administered with a gap of between four to 12 weeks.
