New Delhi, Nov 27 () Biotechnology major Biocon on Wednesday said that US health regulator has approved its and Mylan's supplemental biologics licence application for a drug substance to be manufactured at its new facility in Bengaluru.
 "Biocon and Mylan's supplemental biologics licence application (sBLA)..for pegfilgrastim drug substance to be manufactured in Biocon's new Biologics manufacturing facility has been approved by the US Food and Drug Administration (USFDA)," the company said in a filing to BSE.
 This additional approval of its new manufacturing facility for pegfilgrastim in Bengaluru will enable the biotechnology major to enable its arm Biocon Biologics and Mylan to scale up capacity multi-fold and address the growing market opportunities in US and other global markets, the company said.
 Biocon Biologics CEO Christiane Hamacher said, "This approval will help us better meet global patient needs for Fulphila, a high quality biosimilar pegfilgrastim co developed with Mylan and manufactured by Biocon Biologics".
 Biocon Biologics, through its partner Mylan, has commercialised three of its co-developed biosimilars in developed markets like the US, Canada, the European Union and Australia.
 The USFDA had conducted pre-approval inspection of this new drug substance manufacturing facility from September 10-19, 2019.

Biocon, Mylan get USFDA nod for pegfilgrastim drug substance licence

Biotechnology major Bharat Biotechs BSL-3 (Biosafety Level-3) facilities will play a pivotal role in empowering the company's plans to ramp-up production of India's first indigenously developed Covid-19 vaccine. Accordingly, the biotechnology major plans to produce 700 million doses of <keyword id="9485939" type="PERSON" weightage="20" keywordseo="covaxin">Covaxin</keyword> per annum. The vaccine is regarded as one of the safest in the world and highly efficient against the UK, SA, Brazilian and the new Indian double mutant strain of Covid-19 virus. In a conversation with IANS, Krishna Ella, Chairman and Managing Director of <keyword id="149108" type="PERSON" weightage="20" keywordseo="Bharat-Biotech">Bharat Biotech</keyword> International said the capacity expansion process has been initiated across multiple facilities since early 2021. Presently, the company manufactures 20 million doses per month of Covaxin which are supplied to the <keyword id="210071" type="COMPANY" weightage="20" keywordseo="Ministry-of-Health">Ministry of Health</keyword> for the national vaccine rollout. "Manufacturing scale up has been carried out in a stepwise manner across multiple facilities at Hyderabad, and Bangalore," he said. "Inactivated vaccines, while highly safe, are extremely complex and expensive to manufacture, resulting in lower yields when compared to live virus vaccines." Notably, Bharat Biotech is able to expand Covaxin manufacturing capacity mainly due to the availability of four new specially designed BSL-3 facilities. In industry parlance, a BSL-3 facility is equipped to safely handle infectious biological pathogens in the highest stringent environmentally controlled manufacturing facility. At present, the company has four facilities BSL-3 facilities two in Hyderabad and two in Bengaluru. "These facilities have been repurposed due to a strong pre-existing expertise in development of <keyword id="11129299" type="UNKNOWN" weightage="20" keywordseo="viral-vaccines">viral vaccines</keyword> and know-how applied to manufacture, test and release the highly purified inactivated viral vaccine." The company has recently entered into a 'Master Collaborative Agreement' (<keyword id="6355225" type="COMPANY" weightage="20" keywordseo="mca">MCA</keyword>) with <keyword id="1906647" type="COMPANY" weightage="20" keywordseo="CSIR-Indian-Institute-of-Chemical-Technology">CSIR-Indian Institute of Chemical Technology</keyword> (CSIR-IICT), to collaborate on the development of novel platform technologies in developing a synthetic process route for adjuvant molecule 'TLR 7/8' for Covaxin. Bharat Biotech uses a proprietary adjuvant Algel-IMDG, that has now proven to be a safe and effective adjuvant, especially to stimulate memory T cell responses. The synthesis and manufacture of the <keyword id="11174611" type="COMPANY" weightage="20" keywordseo="imdg">IMDG</keyword> component has been successfully indigenised and will be manufactured at commercial scale within the country. This is the first instance where a novel adjuvant has been commercialised in India. Besides, the company has secured long term components, packing materials and single use consumables supplies for the manufacturing ramp-up To further increase bulk capacity Bharat Biotech has entered into agreements with Indian Immunologicals (IIL) to manufacture the drug substance for Covaxin. "When COVID happened, we said it actually gives us a very strong case to try it (Itolizumab) for COVID, because we do believe that the unique mechanism of action that it has can help us deal with this storm that we have seen is killing patients,'' Mazumdar-Shaw said. Itolizumab is approved for moderate to severe acute respiratory distress syndrome (ARDS) due to COVID-19, she added. On being asked if the company is ramping up the capacity to manufacture more vials on account of the pandemic, she said: "We have the manufacturing capacity and the supply and distribution network in place, however post this approval we are looking to ramp up production capacity to meet the expected surge in demand. Our aim is to reach a larger number of patients across the country". The drug will be manufactured and formulated as an intravenous injection at the company's bio-manufacturing facility at Biocon Park, Bengaluru. New Delhi, April 16 () Pharma and biotechnolgy industry leaders on Thursday stressed on the need for more testing across the country while hailing the government's move to contain the spread of COVID-19 pandemic.This pandemic has put spotlight on science, scientists and biotechnology, Biocon Chairperson Kiran Mazumdar-Shaw said while addressing an online conference organised by the Department of Biotechnology and School of Engineering &amp; Applied Sciences at Bennett University."What we need today is to understand the pandemic and find ways of testing the presence of the virus and ultimately develop a vaccine to counter the virus," she said.It was realised that there is a shortage of testing kits in the country, and there are people who could develop these kits.India has started the manufacturing of indigenous kits to face the challenge of their insufficient availability, Mazumdar-Shaw said in the conference on 'Combating COVID-19: Biotech to the Rescue'.Expressing that the number of tests being done in the country is very small, Sanofi Healthcare Pvt Ltd Chairman Varaprasad Reddy said: "Situation In India is very pathetic regarding the testing. There is lack of test kits. The government has to procure test kits... The <keyword id="43588" type="ORGANIZATION" weightage="20" keywordseo="World-Health-Organization">World Health Organization</keyword> (WHO) special envoy David Nabarro has warned that there is no guarantee that a <keyword id="1754604" type="PERSON" weightage="20" keywordseo="coronavirus">coronavirus</keyword> (COVID-19) <keyword id="24209" type="PERSON" weightage="20" keywordseo="vaccine">vaccine</keyword> can be successfully developed in coming months. Nabarro who is one of the world's leading experts said that people have to live with the threat of coronavirus "for the foreseeable future" and adapt accordingly because there is no guarantee that a vaccine can be successfully developed. In an interview with The Observer, the WHO special envoy said the public should not assume that a vaccine would definitely be developed soon and would have to adapt to the ongoing threat. "You don't necessarily develop a vaccine that is safe and effective against every virus. Some viruses are very, very difficult when it comes to vaccine development - so for the foreseeable future, we are going to have to find ways to go about our lives with this virus as a constant threat," he was quoted as saying. "That means isolating those who show signs of the disease and also their contacts. Older people will have to be protected. In addition, hospital capacity for dealing with cases will have to be ensured. That is going to be the new normal for us all," Nabarro added. Nearly 44 COVID-19 vaccines are currently under various stages of development and scientists say that it will take 12-18 months before the world sees a successful one. According to the World Health Organisation (WHO), 42 vaccines are in pre-clinical stage while only two have entered Phase 1. The two vaccines which are in Phase 1 are from CanSino Biological Inc. and Beijing Institute of Biotechnology; and Moderna/the National Institute of Allergy and Infectious Disease (NIAID) in the US. Pharmaceutical giants Sanofi and GlaxoSmithKline (GSK) have also joined hands to speed up and develop an adjuvanted vaccine for COVID-19 disease. The candidate vaccine is expected to enter clinical trials in the second half of 2020 and, if successful, would be available in the second half of 2021, the two companies said in a statement. As of Sunday morning, the global number of coronavirus cases stood at 2,329,651, with 160,721 deaths.

No guarantee coronavirus vaccine will be developed: WHO envoy

The vaccine NVX-CoV2373 is already being locally manufactured by the company's <keyword id="3658" type="LOCATION" weightage="20" keywordseo="India">India</keyword> partner Pune-based <keyword id="95879" type="COMPANY" weightage="20" keywordseo="Serum-Institute-of-India">Serum Institute of India</keyword> (SII) under the brand name Covovax. In the US-based Phase-3 trials, NVX-CoV2373 also demonstrated 100 per cent protection against moderate and severe disease. "Today, Novavax is one step closer to addressing the critical and persistent global public health need for additional Covid-19 vaccines. These clinical results reinforce that NVX-CoV2373 is extremely effective and offers complete protection against both moderate and severe Covid-19 infection," said Stanley C Erck, President and Chief Executive Officer, Novavax, in a statement. "Novavax continues to work with a sense of urgency to complete our regulatory submissions and deliver this vaccine, built on a well understood and proven platform, to a world that is still in great need of vaccines," Erck added. In August 2020, Novavax and SII announced an agreement under which the US biotechnology company had given SII the licence to manufacture and supply the vaccine in low- and middle-income countries. In March 2021, CEO <keyword id="383467" type="PERSON" weightage="20" keywordseo="Adar-Poonawalla">Adar Poonawalla</keyword> said Covovax will be launched by September this year. "Covovax trials finally begin in India; the vaccine is made through a partnership with Novavax and Serum Institute of India," said Adar Poonawalla. The US-based Phase-3 trials began in December 2020 and enrolled 29,960 participants across 119 sites in the US and Mexico to evaluate efficacy, safety and immunogenicity, with an emphasis on recruiting a representative population of communities and demographic groups most impacted by the disease. The study "confirms that NVX-CoV2373 offers a reassuring tolerability and safety profile", said Gregory M Glenn, MD, President of Research and Development, Novavax. The company will file for regulatory authorisations in the third quarter. After receiving the approvals, Novavax will reach a manufacturing capacity of 100 million doses per month by the end of the third quarter and 150 million doses per month by the end of the fourth quarter of 2021, the statement said. NVX-CoV2373 was created using Novavax' recombinant nanoparticle technology to generate antigen derived from the coronavirus spike (S) protein and is formulated with Matrix-M adjuvant to enhance the immune response and stimulate high levels of neutralising antibodies. It contains purified protein antigen and can neither replicate, nor can it cause Covid-19. The vaccine can be stored at 2 to 8 degrees Celsius, allowing the use of existing vaccine supply chain channels for its distribution. It is packaged in a ready-to-use liquid formulation in 10-dose vials. The two-shot vaccine is also being evaluated in Phase 3 trials in the UK that demonstrated efficacy of 96.4 per cent against the original virus strain, 86.3 per cent against the B117 (Alpha) variant and 89.7 per cent overall. It is also being tested in two ongoing Phase 2 studies that began in August 2020: A Phase 2b trial in South Africa that demonstrated 55 per cent efficacy overall in HIV-negative participants and 48.6 per cent efficacy against the B1351 (Beta) variant, and a Phase 1/2 study in the US and Australia. The company is also conducting trials in adolescents between ages 12 and 18. Moderna will surge another 60% on the value of its potential coronavirus vaccine, according to one Wall Street firm

New Delhi, Jan 6 () Biotechnology major Biocon on Monday said private equity firm True North will invest Rs 536.25 crore to acquire 2.44 per cent stake in its arm Biocon Biologics, valuing the subsidiary at Rs 21,450 crore on a pre-money equity basis.
 The board of Biocon Biologics India Ltd has approved a primary equity investment by True North. As per the terms of the proposed agreement, True North will invest Rs 536.25 crore and will eventually translate to a 2.44 per cent minority stake in the biosimilars business after all restructuring, including the filed merger of Biocon Research Ltd with Biocon Biologics, Biocon said in a filing to the BSE.
 This values Biocon Biologics at Rs 21,450 crore or around USD 3 billion, on a pre-money equity basis, it added.
 "This investment is an endorsement of the value that Biocon Biologics has demonstrated as a global biosimilars player, having commercialised three of its molecules in developed markets such as the EU, the US, Japan and Australia," Biocon Chairperson and MD Kiran Mazumdar-Shaw said.
 This is at an important inflection point as, "we gain commercial success with our first wave of Biosimilar products and invest in the long term through broadening our pipeline, expanding our manufacturing capacity and increasing our commercial footprint," she added.
 In similar vein, Biocon Biologics CEO Christiane Hamacher said the investment by True North in Biocon Biologics reflects a high level of conviction in Biocon Biologics' status as a global frontrunner in biosimilars that is leveraging its large-scale manufacturing capabilities to shift the access paradigm for these life-saving therapies.
 "True North has had a long successful association with Kiran Mazumdar-Shaw and the entire Biocon team and see them as pioneers and leaders in specialised niches," True North partner Satish Chander said.
 True North is happy to associate with them for the third time through this investment in Biocon Biologics. Biosimilars is a fast-growing industry that addresses a fundamental need of bringing affordability to specialised effective biologic products, he added.
 Biocon Biologics has a product pipeline of 28 molecules, including 11 with Mylan, several with Sandoz, and is developing many independently.
 Shares of Biocon Ltd were trading at Rs 287.40 per scrip on the BSE, down 4.53 per cent from its previous close. AKT HRS

The FICM highlighted that many admitted to intensive care with COVID-19 have developed a condition called acute respiratory distress syndrome (ARDS).<a href="https://www.ncbi.nlm.nih.gov/pubmed/32007143" target="_blank" rel="noopener noreferrer">One study published in the US National Center for Biotechnology Information</a> found that 17% of 99 coronavirus patients in Wuhan, China, examined between January 1 and January 20, had developed ARDS during their illness.<a href="https://www.thelancet.com/journals/lancet/article/PIIS0140-6736(20)30183-5/fulltext" target="_blank" rel="noopener noreferrer">Another study published in The Lancet</a> on February 15 found that 29% of 41 patients observed between mid-December to early January in Wuhan had developed ARDS.ARDS prevents a person's lungs from providing their vital organs with enough oxygen, <a href="https://www.nhs.uk/conditions/acute-respiratory-distress-syndrome/">according to the National Health Service</a> (NHS).When COVID-19 reaches the lungs, their mucous membranes - which <a href="https://www.britannica.com/science/mucous-membrane">line</a> the various body cavities and air tracts - become inflamed. According to the NHS, this inflammation can then cause ARDS, whereby "fluid from nearby blood vessels to leak into the tiny air sacs in your lungs, making breathing increasingly difficult."Though the lungs of coronavirus survivors could return to "apparently normal" after six months with minimal issues - like a weakened ability to exercise - those who go on to develop ARDS could "take as long as 15 years for their lungs to recover," the FICM said, according to The Sunday Times.ARDS is responsible for 10% of all intensive care unit admissions in the UK, The Sunday Times said.ARDS also has a mortality rate of 30% to 40%, the newspaper added, citing Michael Matthay, an expert on the disease. This fatality rate was also mentioned in a <a href="https://www.ncbi.nlm.nih.gov/pmc/articles/PMC4456061/">2015 paper</a> on the disease published in the US National Center for Biotechnology Information."There is no specific treatment except to sedate patients and put them on mechanical ventilators to let them recover," Matthay said, referring to ARDS sufferers."Survivors have significant exercise limitation and poor physical quality of life ... related to marked muscle wasting and weakness."ARDS is also commonly found in people recovering from severe flu and pneumonia cases in the UK, but in those cases "it's unusual for ARDS to cause long-term lung damage," <a href="https://www.blf.org.uk/support-for-you/ards">according to the British Lung Foundation</a>.A study <a href="https://jamanetwork.com/journals/jamainternalmedicine/fullarticle/2763184">published March 13 in The Journal of the American Medical Association</a> found that older people with COVID-19 are the most likely group to contract ARDS, "likely owing to less rigorous immune response."The FICM also warned that other organs could also be damaged by COVID-19 after recovery."Like many other viral conditions, the effects of coronavirus are not just limited to the lungs. The heart can also be affected, ranging from inflammation (myocarditis) to heart failure," the FICM said, according to The Sunday Times.Earlier this month the Hong Kong Hospital Authority also found that two to three people out of a group of 12 recovered coronavirus patients were <a href="https://www.businessinsider.com/coronavirus-recovery-damage-lung-function-gasping-air-hong-kong-doctors-2020-3">left with a drop in 20% to 30% in lung function</a>.
